چکیده
Abstract Anti-citrullinated protein antibodies (ACPAs) are a hallmark of rheumatoid arthritis, but the sources citrullinated antigens remain incompletely defined. Neutrophil extracellular traps (NETs) appear to be source ACPA targets. However, while citrullinating enzyme peptidylarginine deiminase (PAD)4 is required for producing NETs, it not citrullination in murine arthritis. Conversely, PAD2 has limited role NETs major contributor proteins. Thus, additional likely exist. We investigated if macrophage (METs) could proteins or PAD4 dependent manner. Thioglycolate induced macrophages from wild-type, −/−and −/−mice were activated with variety stimulants, then fixed and stained DAPI either anti-citrullinated histone H4 (H4C3) antibody sera ACPA+ ACPA− arthritis subjects. METs visualized by immunofluorescence, confirmed using DNase, counted blinded manner, compared paired-T test. found that ionomycin monosodium urate (MSU) crystals induce (H4C3 stained). In absence PAD4, reduced response MSU when wildtype, complete −/−macrophages. Also, IgG sera, bound METs. Finally, MSU. contribute generation targeted ACPAs PAD2− PAD4-dependent Funding this work was provided U.S. Army MedicalResearch Acquisition Activity through Peer Reviewed Program (W81XWH-18-1-0717) M.A.S.

Localization of peptidylarginine deiminase 4 (PADI4) and citrullinated protein in synovial tissue of rheumatoid arthritis.
OBJECTIVES Peptidylarginine deiminases (PADIs) convert peptidylarginine into citrulline via post-translational modification. Anti-citrullinated peptide antibodies are highly specific for rheumatoid arthritis (RA). Our genome-wide case-control study of single-nucleotide polymorphisms found that the PADI4 gene polymorphism is closely associated with RA. Here, we localized the expression of PADI4 ...
متن کاملSmoking is not linked to the development of anti-peptidylarginine deiminase 4 autoantibodies in rheumatoid arthritis
BACKGROUND Defining environmental factors responsible for development of autoimmunity in rheumatoid arthritis (RA) is critical for understanding mechanisms of disease initiation and propagation. Notably, a history of cigarette smoking has been implicated in the genesis of RA and is associated with worse disease outcomes. Antibodies to peptidylarginine deiminase 4 (PAD4) are also associated with...

The discovery that citrullination was crucial for the recognition of antigens by the most disease-specific class of autoantibodies in rheumatoid arthritis (RA) had a huge impact on studies aimed at understanding autoimmunity in this disease. In addition to the detailed characterization of anti-citrullinated protein antibodies, various studies have addressed the identity of citrullinated antigen...

there are controversial reports regarding the role of peptidylarginine deiminase type 4 (padi4) gene polymorphisms and risk of rheumatoid arthritis (ra). the aim of the present study was to investigate the impact of padi4 rs1748033 polymorphism and susceptibility to ra in a sample of the iranian population.this case-control study was done on 150 patients with ra and 150 healthy subjects.padi4 r...
متن کاملPotential protein targets of the peptidylarginine deiminase 2 and peptidylarginine deiminase 4 enzymes in rheumatoid synovial tissue and its possible meaning.
OBJECTIVE The molecular mechanism of citrullination involves the calcium-dependent peptidylarginine deiminase (PAD) family of enzymes. These enzymes induce a stereochemical modification of normal proteins and transform them into autoantigens, which in rheumatoid arthritis trigger a complex cascade of joint inflammatory events followed by chronic synovitis, pannus formation, and finally, cartila...
